Analytical Method Development and Validation for Simultaneous Estimation of Paracetamol and Piroxicam by UV Method

Abstract

A new establishment and validation of a simple, precise, accurate and cost effective UV-Visible spectrophotometric method was developed to estimate both Paracetamol and Piroxicam simultaneously in conjoined pharmaceutical dosage forms. This technique was founded on simultaneous equation method of absorbances recorded at 252.0 nm and 282.0 nm of Paracetamol and Piroxicam in methanol respectively and an isosbestic point could be observed at 265.0 nm. It showed very good linearity over concentration range of 5 30 UV/mL of both drugs with correlation coefficient (R 2 ) of 0.997 to Paracetamol and 0.992 to Piroxicam. Repeatability and reproducibility were ensured due to results of the precision studies with %RSD values that were within the acceptable range. The recovery experiments confirmed the accuracy and gave the values of recovery Paracetamol =78.9 and Piroxicam=88.6. The approach was harsh and hardy when there would be slight strained changes in the lab oratory conditions. The determined values of LOD and LOQ were 60.46 5g/mL and 194.32 5g/mL of Paracetamol and 57.98 5g/mL and 175.71 5g/mL of Piroxicam, respectively. The adopted procedure is easy, fast and reproducible in daily simultaneous determination of Paracetamol and Piroxicam in bulk drugs and co-formulated tablets, in accordance with ICH Q2 (R1) regulations.
