Abstract
Diacerein, an anti-inflammatory drug, which comes under BCS class II, commonly used in treatment of osteoarthritis has limited by its poor solubility and bioavailability when administered topically. This study aimed to develop and evaluate diacerein-loaded nanosponges gel using varying concentrations of ethyl cellulose and polyvinyl alcohol (PVA) to enhance its delivery. Nanosponges were prepared using an emulsion solvent evaporation technique, with different ratios of ethyl cellulose (0.05-0.250%) and PVA (0.2-0.3%). The formulations were evaluated for production yield, entrapment efficiency, Fourier Transform Infrared Spectroscopy (FTIR) analysis, and in vitro drug release. Further the prepared nanosponges are incorporated into the gel base prepared by two different polymer carbopol-934 and carbopol-940 the prepared gels were further evaluated for viscosity, spreadability, drug content and in-vitro studies. Among the formulations tested, F10 exhibited the optimal characteristics with a particle size of 343.2nm. Diffusion studies indicated a significant increase in diacerein penetration compared to control formulations and there was no skin irritation found in rats. The diacerein-loaded nanosponges gel, particularly formulation F10, demonstrated promising potential for enhanced topical delivery of diacerein. This formulation may provide a more effective treatment option for inflammatory conditions, warranting further clinical evaluation.
