Abstract

Adipose tissue–derived mesenchymal stem cells (AT-MSC) represent a heterogeneous population of self-renewing, multipotent progenitor cells residing in the stromal compartment of adipose tissue. Canine adipose tissue-derived mesenchymal stem cells have attracted significant interest in regenerative medicine due to their therapeutic potential in naturally occurring canine diseases and their translational relevance to human medicine. In the present study, canine adipose tissue–derived MSCs (cAT-MSCs) were isolated and differentiated into osteogenic, chondrogenic and adipogenic lineages. Their immunomodulatory gene expression profiles were examined to evaluate their immunomodulatory capacity. cAT-MSCs were isolated and characterized by morphology, molecular profiling (CD44, CD73, CD90, CD105 positive; CD45 negative), and trilineage differentiation. Gene expression analysis demonstrated the presence of transcripts encoding vascular endothelial growth factor (VEGF), interleukin (IL)-6 and tissue inhibitors of metalloproteinases-2 (TIMP2, whereas IL-10, TGF-β, COX2, CCL2, IDO transcripts were not detected. Collectively, these findings indicate that the immunomodulatory activity of cAT-MSCs is primarily mediated through soluble factors such as IL6, VEGF and TIMP2. The results support the potential application of cAD-MSCs as a therapeutic tool for the management of immune-mediated disorders.
