Abstract

Now a day’s researcher’s curiosity about gastro-retentive drug delivery system (GRDDS) increased so high in recent years because of its excellent drug releasing properties to the target site. The delivery system’s residence time in the stomach can be extended with the usage of GRDDS by using a number of natural and synthetic polymers like guar gum, HPMC (Hydoxy Propyl Methyl Cellulose) which can be used to improve the bioavailability of a certain pharmacological ingredient by releasing the medication at a specific absorption site also improved gastric retention time of drug, hence overcoming the limitations associated with standard oral dose forms. Drug solubility is increased by prolonging the retention of medications in the stomach, especially those with low solubility at high intestinal pH. Both local and systemic effects of GRDDS have been demonstrated to be beneficial in the treatment of duodenal or stomach ulcers. By releasing the medication locally, GRDFs significantly enhance stomach pharmacotherapy by resulting in a high concentration of the drug at the gastric mucosa which has the potential to last for an extended period of time. GRDFs provide continuous and sustained drug release to the upper portion of the gastrointestinal tract (GIT) by lengthening the period of drug release and enhancing the bioavailability of drugs with a small therapeutic window. This prolongs the interval between doses and improves patient compliance. This review aims to provide a brief overview of gastro retentive drug delivery (GRDD), including its associated variables, benefits, and drawbacks. It emphasizes the importance of GRDD over traditional method of administering medication.
