Special Issue Description

Lysosomes are membrane-enclosed organelles that contain an array of enzymes capable of breaking down all types of biological polymers—proteins, nucleic acids, carbohydrates, and lipids. The outflow of the byproducts of intralysosomal catabolism is facilitated by their physiological function. With rare exceptions, these porters are uncommon candidates for the translocation of xenobiotics across the lysosome membrane due to their specialization. Passive diffusion is most likely to be used to achieve efflux from the lysosome where it is possible. Experimental investigations of passive diffusion over the lysosome membrane have revealed that it shares many properties with other biological membranes. When drug delivery options are considered that involve a drug-conjugate reaching the lysosome compartment and drug release there by the lysosomal enzymes, predictions of how easily molecules will pass through lysosomal membranes are valuable. These methods won’t work if the medication can’t exit the lysosome and get to the cells where it needs to act pharmacologically.
