By observing the surface plasmon resonance (SPR) band shift of gold nanoparticles with some diameters, the adsorption of the SARS-CoV-2 Omicron spike protein to the nano-gold colloid surfaces was investigated. The reversible creation of the gold colloid aggregates was triggered by the externally altered pH between 3 and 11 at 24.5 0.4 °C. The formation/deformation of the aggregates was observed as a red/blue shift in the peak of the SPR band. For the gold colloids with d = 10, 15, and 20 nm, there was no evidence of reversible aggregation. The coronavirus, or SARS-CoV-2, has experienced spontaneous mutation and antigenic variation throughout time and numerous extremely alarming forms, including the Alpha (B.1.1.7), Beta (B.1.351), Gamma (P.1), and Delta (B.1.617.2) coronaviruses, have been described. The spike protein has 32 mutations in the Omicron variation (B.1.1.529), which significantly increased its pathogenicity when compared to earlier variants. Omicron has a 5.5–11 times greater mutation rate in the receptor-binding motif (RBM) than Alpha, Beta, and Delta SARS-CoV-2 variants. The receptor binding domain (RBD) of the S1 subunit spike protein is made up of 319–541 acids. G339D, S371L, S373P, S375F, K417N, N440K, G446S, S477N, T478K, E484A, Q493R, G496S, Q498R, N501Y, and Y505H are the reported mutations for Omicron in the RBD region.
