Special Issue Description

The determination of pharmacological targets is a crucial phase in the drug design process in the biomedical field. Therefore, it is tempting to also use this model to describe genetic diseases. Although the model works well for genetic disorders with gain-of-function, it is less effective for the majority of genetic diseases with loss of function. The majority of common diseases, along with hereditary diseases with gain-of-function, are defined by the activation of particular pathways or the ectopic activity of proteins, which make excellent targets. On the other hand, loss-of-function genetic illnesses are brought on by the deficiency of a single protein and may have widespread effects. It may be challenging to identify pharmaceutically relevant targets for such disorders since the concept of a pharmacological target is less exact. To prevent allocating resources to ineffective techniques in the search for cures, it is important to take into account this important but usually overlooked element of loss-of-function genetic illnesses.
