    Osteopontin (OPN), a calcified ECM-associated protein similar to a chemokine, plays a key role in determining the metastatic potential of several cancers. Extensive research from several laboratories has demonstrated the likely role of OPN in regulating cell signaling that ultimately controls tumor growth and metastasis. It can function as both a cell attachment protein and a cytokine, delivering signals to cells via a range of receptors including several integrins and CD44. OPN expression is increased by a variety of toxins, especially those that activate protein kinase C.

    As a signaling molecule, OPN can alter gene expression and promote monocyte/macrophage migration up an OPN gradient. It has both inflammatory and anti-inflammatory effects. Some experiments suggest that it can inhibit apoptosis, potentially aiding cell survival in response to toxic damage. The increase in OPN expression is often related to malignancy and has been shown to improve the tumorigenic and/or metastatic phenotype of the cancer cell.
